Studies on the methodology of 18F-FDG cell binding of ovarian cancer SKOV3

Abstract

Objective: To establish a useful and stable methodology of 18F-FDG cell binding of ovarian cancer SKOV3. Methods: Several binding conditions were tested. ①The cell number was from 5×104 to 5×106 cells/bottle. ②The reaction time was from 20 to 120min. ③The radioactivity of 18F-FDG was from 1.85 to 29.6KBq. ④The glucose concentration was from 0 to 11.1mmol/L. The CPM(B) in cells and CPM(F) in supernatant were measured by the γ detector. The binding efficiency of ovarian cancer cell line SKOV3 with 18F-FDG was calculated. Results: ①There was linear regression relationship between 18F-FDG cell binding efficiency of human ovarian cancer SKOV3(y) and the cell number(x1), reaction time(x2), glucose concentration(x3). Regression equation: y=(7.395+5.18E-0.06x1+0.094x2-2.040x3)×100%, F=106.9, P<0.05. The P values of partial regression coefficient were less than 0.05. R2c=0.728, P<0.05. The Pearson correlation coefficient of cell number, reaction time, glucose concentration with 18F-FDG cell binding efficiency was 0.581、0.179、-0.599 respectively, P<0.05. ②The basic conditions of 18F-FDG cell binding of ovarian cancer SKOV3: the cell number was 1×106 cells/bottle, the radioactivity of 18F-FDG was 3.7KBq, the glucose concentration was 0mmol/L, the reaction time was 100min. The binding efficiency was (25.45±1.66)%. Conclusions: The methodology of 18F-FDG cell binding of ovarian cancer SKOV3 was established successfully. It contributed to evaluate early the effect of radiotherapy and chemotherapy on inhibiting ovarian cancer cell line SKOV3.
